PADRAIC

PADRAIC is a suite of studies, in the main funded by the CRUK Leeds-Manchester PhD Collaboration, administrated through the Manchester Cancer Research Centre (MCRC), and based between the Obesity-Cancer Research Group (lead: Renehan) and the Leeds CTRU (Leads: Brown & Wilson).

The studies include a series of systematic reviews and meta-analyses to (i) quantify effects sizes from various anti-obesity interventions (PADRAIC-1, -2, -3); (ii) quantify cancer risk reduction effects from various anti-obesity interventions (PARDAIC-4, -5); (iii) an international expert consensus on the need for an design of a large scale weight loss to prevent cancer clinical trial (PADRAIC-6), and (iv) an expert elicitation study (using the SHELF framework) to elicit best estimates of cancer risk reduction for a given weight loss. The results from these studies will populate an In Silico Bayesian probability simulation model (PADRAIC-7) that will serve as a platform to estimate sample sizes, and uncertainties, for a long-term cancer prevention trial using anti-obesity interventions (behavioural, drug-based, bariatric surgery-based). 

How it works

PADRAIC projects

PADRAIC is a suite of studies aiming to understand the feasibility of a large-scale cancer prevention intervention. Currently there are 8 studies, which are outlined below. More information for each study can be found on the individual webpages.

PADRAIC 1-3

These three studies aim to understand the efficacy of weight loss of different weight loss interventions.

They also aim to understand the variability in adherence and follow up and the reporting of socio-economic and ethnic status

PADRAIC 4-5

Studies 4 and 5 are systematic review and meta analysis aiming to understand the impact of GLP-1 receptor agonist on cancer risk and behavioural intervention on cancer risk.

PADRAIC 6

An international expert consensus was undertaken to understand the need for a clinical trial INVESTIGATING weight loss intervention and cancer prevention.

PADRAIC 7-8

PADRAIC 7 is an expert elicitation of a probability distribution key to in-silico trial design.

This elicited parameter will be used to creat an in-silico trial model of weight loss INTERVENTION and its impact on cancer risk. This model will allow us to estimate samlpe sizes given key uncertainty and possible designs.

PADRAIC is funded by Cancer Research UK as a Leeds-Manchester collaborative clinical research training fellow grant.
